目的研究胃癌和癌旁组织中Aurka mRNA及其蛋白的表达,并分析胃癌组织中Aurka mRNA表达水平与患者临床病理学特征的关系。方法回顾性收集2011年4月至2013年9月期间于川北医学院附属医院胃肠外科接受手术的198例胃癌患者的手术标本,包括癌组织和癌旁组织。采用实时荧光定量逆转录聚合酶链反应(RT-PCR)和免疫组织化学染色法分别检测胃癌和癌旁组织中Aurka mRNA及其蛋白的表达,并进行比较;同时探讨胃癌组织中Aurka mRNA表达水平与患者临床病理学特征的关系。结果 RT-PCR结果显示,胃癌组织与癌旁组织中Aurka mRNA的表达水平分别为16.62±1.85和7.10±1.59,癌组织较高,差异有统计学意义(t=-38.58,P<0.05);免疫组织化学染色结果显示,胃癌组织中Aurka蛋白的表达阳性率高于癌旁组织〔93.9%(186/198)比16.2%(32/198),P<0.01〕。胃癌组织中Aurka mRNA的表达水平与性别、年龄、肿瘤位置、术前癌胚抗原(CEA)和CA19-9表达均无关(P>0.05),而与肿瘤TNM分期、T分期、N分期及分化程度均有关(P<0.05)。幽门螺杆菌(HP)阳性患者胃癌组织中Aurka mRNA的表达水平为15.38±1.73,高于HP阴性患者(7.20±1.86),差异有统计学意义(t=-3.74,P<0.01)。结论 Aurka基因及其蛋白在胃癌组织中的表达水平均高于癌旁组织,提示其作为一种致癌基因,在胃癌的发生和发展过程中扮演着重要角色,可作为胃癌患者预后及复发的预测指标。
Objective To investigate the expression of Aurka mRNA and its protein in gastric cancer and its adjacent tissues and to analyze the relationship between Aurka mRNA expression and clinicopathological features in gastric cancer. Methods Retrospectively collected surgical specimens from 198 patients with gastric cancer who underwent surgery in the Department of Gastrointestinal Surgery, Affiliated Hospital of North Sichuan Medical College from April 2011 to September 2013, including cancer tissues and paracancerous tissues. The expression of Aurka mRNA and its protein in gastric cancer and paracancerous tissues were detected by real-time fluorescent quantitative reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemical staining, respectively. Results The expression of Aurka mRNA in gastric cancer tissues and adjacent tissues was 16.62 ± 1.85 and 7.10 ± 1.59, respectively. There was a significant difference between the two groups (t = -38.58, P <0.05). Immunohistochemical staining showed that the positive expression rate of Aurka protein in gastric cancer tissues was higher than that in adjacent tissues [93.9% (186/198) vs 16.2% (32/198), P <0.01). The expression level of Aurka mRNA in gastric cancer tissues was not correlated with gender, age, tumor location, preoperative carcinoembryonic antigen (CEA) and CA19-9 (P> 0.05), but not with tumor TNM stage, T stage, N stage and differentiation Degree (P <0.05). The expression level of Aurka mRNA in gastric cancer tissue of Helicobacter pylori (HP) positive patients was 15.38 ± 1.73, higher than that of HP negative patients (7.20 ± 1.86), the difference was statistically significant (t = -3.74, P <0.01). Conclusion The expression level of Aurka gene and its protein in gastric cancer tissues is higher than that in paracancerous tissues, suggesting that Aurka gene and its protein play an important role in the carcinogenesis and progression of gastric cancer and can be used as a prediction for the prognosis and recurrence of gastric cancer index.
