目的研究两种黏细菌代谢产物(NX52和NX83)对人结直肠癌细胞增殖的影响,探讨其可能的作用机制。方法用不同浓度(0.1 mg/m L、1.0 mg/m L和10.0 mg/m L)的黏细菌代谢产物分别处理结直肠癌细胞HT-29、SW480和SW1463,阴性对照组的肿瘤细胞不加黏细菌代谢产物,采用四甲基偶氮唑蓝比色法检测其对结直肠癌细胞的增殖抑制效应,采用细胞形态学及流式细胞术观察细胞凋亡情况。结果两种代谢产物NX52及NX83均对人结直肠癌细胞HT-29、SW480和SW1463的细胞增殖有抑制作用(P<0.05),且呈时间剂量效应;代谢产物NX83对肿瘤细胞增殖抑制作用较NX52更明显(P<0.05);与阴性对照组相比,代谢产物NX83作用SW1463细胞48 h后凋亡率明显增加(P<0.01)。结论黏细菌代谢产物NX52和NX83具有体外抗癌活性,作用机制可能是通过诱导细胞凋亡杀伤肿瘤细胞。
Objective To study the effects of two myxobacteria metabolites (NX52 and NX83) on the proliferation of human colorectal cancer cells and to explore their possible mechanism. Methods Colorectal cancer cells HT-29, SW480 and SW1463 were treated with myxobacteria metabolites of different concentrations (0.1 mg / mL, 1.0 mg / mL and 10.0 mg / mL respectively), and the tumor cells in the negative control group Myxo bacteria metabolites, the use of tetramethylzirconyl blue colorimetric assay of colorectal cancer cell proliferation inhibitory effect, the use of cell morphology and flow cytometry observation of apoptosis. Results The two metabolites, NX52 and NX83, both inhibited the proliferation of human colorectal cancer cells HT-29, SW480 and SW1463 (P <0.05), and showed a time-dose effect. The metabolite NX83 inhibited the proliferation of tumor cells (P <0.05). Compared with the negative control group, the apoptosis rate of SW1463 cells treated with the metabolite NX83 was significantly increased (P <0.01). Conclusions Myxo-bacteria metabolites NX52 and NX83 have antitumor activity in vitro. The mechanism may be that tumor cells are killed by inducing apoptosis.
