目的:观察葛根素对急性肺血栓栓塞溶栓治疗后P、E-选择素表达的影响。方法:24只日本大耳白兔,随机分为假手术组(S组)、单纯溶栓组(T组)、葛根素组(Pur组),经右心导管注入条柱状自体血栓,心导管法监测血流动力学,制备急性肺血栓栓塞实验模型。测定栓塞前、溶栓前即刻、溶栓后2h、4h血浆中超氧化物歧化酶(SOD)、丙二醛(MDA)的含量;采用免疫组化及原位杂交等方法观察肺微血管P、E-选择素表达的变化。结果:(1)T组和Pur组在溶栓开始后1h,PAMP即显著下降(P<0.01);在溶栓开始后1h Pur组PAMP即明显低于T组(P<0.05)。(2)与T组同期比较,Pur组在溶栓后2h、4h血浆SOD活力和MDA含量分别有明显升高和下降(P<0.05,P<0.01)。(3)Pur组与T组比较肺微血管P、E-选择素和P、E-选择素mRNA平均吸光度(А)值均有明显下降(P<0.05)。结论:葛根素可下调急性肺血栓栓塞溶栓后肺微血管P、E-选择素的表达,对溶栓治疗后再灌注损伤有保护作用。
Objective: To observe the effect of puerarin on the expression of P and E-selectin in patients with acute pulmonary thromboembolism after thrombolysis. Methods: Twenty-four Japanese white rabbits were randomly divided into sham-operation group (S group), simple thrombolysis group (T group) and Puerarin group (Pur group) Method to monitor hemodynamics, the preparation of experimental model of acute pulmonary thromboembolism. The levels of plasma SOD and MDA were measured before embolization, immediately before thrombolysis, 2h and 4h after thrombolysis. The expressions of P and E were observed by immunohistochemistry and in situ hybridization - Changes in the expression of selectins. Results: (1) PAMP was significantly decreased at 1 hour after initiation of thrombolysis in both T and Pur groups (P <0.01). PAMP in Pur group was significantly lower than that in T group at 1 hour after thrombolysis (P <0.05). (2) Compared with the same period of T group, plasma SOD activity and MDA content in Pur group were significantly increased and decreased at 2 and 4 hours after thrombolysis (P <0.05, P <0.01). (3) Compared with T group, the average absorbance (P) of pulmonary microvascular P, E-selectin and P, E-selectin mRNA in the Pur group were significantly decreased (P <0.05). CONCLUSION: Puerarin can down-regulate the expression of P and E-selectin in pulmonary microvascular after acute pulmonary thromboembolism thrombolysis, and has a protective effect on reperfusion injury after thrombolytic therapy.
