本文用酶联免疫吸附试验(ELISA)间接法测定粉尘螨(Dermatophagoides frinae)特异性IgE和IgG,双抗体法测定总IgE。实验中采取棋盘滴定,选择了包被的抗原或抗体、血清和酶结合物的最适浓度,并对抗原包被条件、血清和酶结合物保温条件等作了比较,经抑制试验、平行性和重复性试验,认为ELISA的特异性、敏感性和重复性均较好,而且操作简易、快速。测定总IgE的敏感范围在2～60IU/ml。本文还对影响实验的因素作了讨论。
In this paper, Dermatophagoides frinae specific IgE and IgG were determined by indirect enzyme-linked immunosorbent assay (ELISA) and total IgE by double antibody method. In the experiment, we took checkerboard titration and chose the optimal concentration of coated antigen or antibody, serum and enzyme conjugate, and compared the antigen coating conditions, the incubation conditions of serum and enzyme conjugate. After the inhibition test, the parallelism And repeatability test, that ELISA specificity, sensitivity and repeatability are better, and easy to operate, fast. Sensitive range of determination of total IgE in 2 ~ 60IU / ml. This article also discusses the factors that affect the experiment.
