解毒化瘀生津方对干燥综合征模型小鼠颌下腺IFN-γ、IL-4影响

目的 :观察解毒化瘀生津方对干燥综合征小鼠颌下腺干扰素-γ(Interferon-γ,IFN-γ)、白介素-4(Interleukin-4,IL-4)蛋白含量表达的影响,探讨其作用机制。方法 :采用免疫诱导法建立干燥综合征小鼠动物模型,以解毒化瘀生津方灌胃治疗,并以硫酸羟氯喹片(Hydroxychloroquine Sulfate,HCQ)灌胃作为对照,采用酶联免疫吸附法检测颌下腺IFN-γ、IL-4蛋白含量表达,并对小鼠颌下腺IFN-γ、IL-4进行免疫组织化学法检测及免疫荧光染色观察。结果 :酶联免疫吸附法检测发现,模型组颌下腺IFN-γ、IL-4的蛋白含量及IFN-γ/IL-4比值高于正常组(P<0.05,P<0.01),中药中、高剂量组和硫酸羟氯喹组IFN-γ、IL-4蛋白含量及IFN-γ/IL-4比值低于中药低剂量组(P<0.05)。免疫组化方法检测小鼠颌下腺IFN-γ、IL-4的平均吸光度,模型组颌下腺IFN-γ、IL-4的吸光度高于正常组(P<0.05),中药中、高剂量组和硫酸羟氯喹组吸光度低于中药低剂量组(P<0.05),接近于正常组。免疫荧光技术观察,模型组荧光染色最强,正常组染色最弱。结论 :解毒化瘀生津方能降低干燥综合征模型小鼠颌下腺IFN-γ、IL-4蛋白含量的表达,恢复了Th1/Th2的平衡稳态,减轻颌下腺的病理损伤,与硫酸羟氯喹的作用相当。 Objective: To observe the effects of Jiedu Huayu Shengjin Fang on protein expression of Interferon-γ (IFN-γ) and Interleukin-4 (IL-4) in the mice with Sjogren’s Syndrome mechanism. Methods: Animal model of Sjogren’s syndrome mice was established by immune induction method, and treated with detoxification, blood stasis and salvia miltiorrhiza recipe. Ginseng was administrated with Hydroxychloroquine Sulfate (HCQ) as the control, and the submandibular gland was detected by enzyme-linked immunosorbent assay IFN-γ, IL-4 protein expression, and immunohistochemistry and immunofluorescent staining of IFN-γ, IL-4 in submandibular gland of mice. Results: The levels of IFN-γ and IL-4 in submaxillary gland of model group were significantly higher than those in normal group (P <0.05, P <0.01) The content of IFN-γ, IL-4 and the ratio of IFN-γ / IL-4 in the dose group and the hydroxychloroquine group were lower than those in the low dose group (P <0.05). The mean absorbance of IFN-γ and IL-4 in submandibular gland of mice was detected by immunohistochemistry. The absorbance of IFN-γ and IL-4 in submaxillary gland of model group was higher than that of normal group (P <0.05) Chloroquine group absorbance lower than the traditional Chinese medicine low dose group (P <0.05), close to the normal group. Immunofluorescence staining showed that the model group had the strongest staining and the normal group had the weakest staining. Conclusion: Jiedu Huayu Shengjin can reduce the expression of IFN-γ and IL-4 in the submandibular gland of mice with Sjogren’s syndrome, restore the balance homeostasis of Th1 / Th2, alleviate the pathological damage of submandibular gland, and the effect of hydroxychloroquine sulfate quite.
