目的:研究不同安络小皮伞菌丝体中粗多糖的理化性质及单糖组成,测定2种安络小皮伞菌总糖、酸性糖和蛋白质含量,并计算单糖组成摩尔比。方法:采用水提醇沉法提取2种安络小皮伞多糖,通过苯酚-硫酸比色法测定多糖提取部位的总糖含量,用间羟基联苯法测定酸性糖含量,用Lowry法测定蛋白质含量,柱前衍生化HPLC法测定单糖组成。结果:固体发酵安络小皮伞、液体发酵小皮伞总糖含量分别为46.78%、26.08%,蛋白质含量分别为30.84%、46.23%,酸性糖含量均不高。2种安络小皮伞均由甘露糖、鼠李糖、葡萄糖、半乳糖、木糖、阿拉伯糖6种单糖组成,固体发酵小皮伞摩尔比为0.051:0.012:1.000:0.070:0.364:0.228,液体发酵安络小皮伞摩尔比为0.108:0.067:1.000:0.221:0.012:0.029。结论:安络小皮伞菌中主要成分为糖肽类物质,二者总量在70%以上,酸性糖含量不高。PMP-HPLC法操作简便,分离效果好,稳定。
OBJECTIVE: To study the physicochemical properties and the monosaccharide composition of crude polysaccharides in the mycelia of Anlainense parachutera, and to determine the total sugar, acid sugar and protein content of two kinds of Acacia punctatus, and to calculate the molar ratio of monosaccharide composition. Methods: Two kinds of polysaccharides were obtained by water extraction and alcohol precipitation. The content of total sugar was determined by phenol-sulfuric acid colorimetric method. The content of acid sugar was determined by the method of m-hydroxybiphenyl. The content of protein was determined by Lowry method Content, pre-column derivatization HPLC determination of monosaccharide composition. Results: The results showed that the total sugar content of liquid fermentation was about 46.78% and 26.08%, respectively, and the content of protein was 30.84% and 46.23% respectively, while the content of acid sugar was not high. The two kinds of anchorage parapsilles consisted of six kinds of monosaccharides, mannose, rhamnose, glucose, galactose, xylose and arabinose. The molar ratio of solid fermentation was 1.51: 0.012: 1.000: 0.070: 0.364: 0.228, and the molar ratio of liquid fermentations to parachutes 0.108: 0.067: 1.000: 0.221: 0.012: 0.029. CONCLUSION: The main component of Agrocybe aegerita is glycopeptide, the total amount of which is above 70% and the content of acid sugar is not high. PMP-HPLC method easy to operate, the separation effect is good and stable.
