目的观察淋病奈瑟菌表面蛋白A(Neisseria surface protein A,NspA)基因真核表达质粒不同免疫途径诱导的小鼠特异性体液免疫应答效果。方法将NspA基因真核表达质粒pcNspA通过肌肉、滴鼻和阴道3种途径免疫BALB/c小鼠,分别于0、2、3、4周各免疫1次,末次免疫后2周,收集各组小鼠血清、支气管肺泡洗液和阴道洗液,采用间接ELISA方法检测特异性抗体效价;并检测抗体介导的补体溶菌活性。结果滴鼻免疫和肌肉免疫pcNspA质粒均可诱导小鼠体内产生较高水平的特异性IgG和IgA抗体,而且滴鼻免疫组在支气管肺泡洗液和阴道洗液中均可检测到特异性sIgA抗体,而阴道免疫组仅在阴道局部检测到较低水平的特异性sIgA抗体;滴鼻免疫组小鼠血清具有溶菌活性。结论淋病奈瑟菌NspA基因真核表达质粒可诱导小鼠体内产生特异性抗体,并具有抗菌活性;滴鼻免疫可能是其更有效的免疫途径。
Objective To observe the effect of specific humoral immune response induced by different immunization routes of Neisseria surface protein A (NspA) gene eukaryotic expression plasmid. Methods The eukaryotic expression plasmid pcNspA of NspA gene was immunized to BALB / c mice by intramuscular, intranasal and vaginal routes. The immunized mice were immunized with 0, 2, 3 and 4 weeks respectively and 2 weeks after the last immunization. Mouse serum, bronchoalveolar lavage fluid and vaginal lotion, and the specific antibody titers were detected by indirect ELISA; and the antibody-mediated complement lysis activity was detected. Results Both intranasal and muscle immunized pcNspA plasmids induced high levels of specific IgG and IgA antibodies in mice, and the specific sIgA antibodies were detected in the BALF and vaginal washings , While the vaginal immunization group only detected a lower level of specific sIgA antibody in the vagina; the serum of the immunized mice had bacteriolytic activity. Conclusion Neisseria gonorrhoeae NspA gene eukaryotic expression plasmid can induce specific antibodies in mice and has antibacterial activity. Intranasal immunization may be a more effective immunization pathway.
