目的探讨髓磷脂碱性蛋白MBP刺激的PBMC培养上清作用的人脐静脉内皮细胞HUVEC对T细胞增殖的影响。方法用细胞免疫化学染色和FCM检测HUVEC的VCAM1ICAM1和HLADR的表达用MTT法测定MBP刺激的PBMC对T细胞增殖的影响。结果与对照相比较MBP刺激的PBMC培养上清可诱导HUVEC特异表达VCAM1和HLADR上调ICAM1的表达。经MBP作用的HUVEC可明显促进T细胞的增殖。结论MBP刺激PBMC的培养上清可引起内皮细胞结构和功能改变促进T细胞增殖。
Objective To investigate the effect of human umbilical vein endothelial cells (HUVECs) stimulated by myelin basic protein MBP on the proliferation of T cells. Methods Immunocytochemical staining and FCM were used to detect the expression of VCAM1ICAM1 and HLADR in HUVECs. The effects of MBP-stimulated PBMC on the proliferation of T cells were determined by MTT assay. Results Compared with the control MBP stimulated PBMC culture supernatant can induce HUVEC-specific expression of VCAM 1 and HLA DR up-regulation of ICAM-1 expression. HUVEC treated with MBP can obviously promote the proliferation of T cells. Conclusion MBP can stimulate the culture supernatant of PBMC to induce changes of endothelial cell structure and function and promote T cell proliferation.
