目的探讨环氧合酶2抑制剂NS398对人卵巢癌细胞系CAOV3细胞周期的影响。方法2003年9月至2004年6月辽宁省肿瘤医院等采用四甲基偶氮唑蓝(MTT)法观察NS398对CAOV3的增殖抑制情况;流式细胞仪测定NS398对CAOV3细胞周期的影响;westernbloting法检测NS398作用前后的细胞周期蛋白cyclinD1、cyclinE的变化。结果NS398对CAOV3细胞的增殖具有抑制作用,并显示出明显的时间和剂量的依赖性;NS398使CAOV3细胞发生细胞周期的G1期阻滞,并抑制细胞周期蛋白cyclinD1、cyclinE的表达。结论NS398可能至少通过抑制cyclinD1和cyclinE的途径抑制CAOV3细胞的增殖。
Objective To investigate the effect of cyclooxygenase 2 inhibitor NS398 on the cell cycle of human ovarian cancer cell line CAOV3. Methods From September 2003 to June 2004, Liaoning Provincial Tumor Hospital et al. (MTT) method was used to observe the inhibitory effect of NS398 on the proliferation of CAOV3 cells. Flow cytometry was used to determine the effect of NS398 on the cell cycle of CAOV3. Westernbloting The changes of cyclin D1 and cyclinE before and after NS398 treatment were assayed. Results NS398 inhibited the proliferation of CAOV3 cells and showed a time-and dose-dependent manner. NS398 blocked the G1 phase of the cell cycle and inhibited the expression of cyclin D1 and cyclinE in CAOV3 cells. Conclusion NS398 may inhibit the proliferation of CAOV3 cells at least by inhibiting cyclinD1 and cyclinE pathways.
