目的探讨三七总皂苷对阿霉素肾病大鼠肾组织内皮素-1蛋白及mRNA表达的影响。方法将45只大鼠随机分为5组,正常组、假手术组、模型组、苯那普利组和三七总皂苷组,手术切除大鼠右侧肾脏并腹腔注射阿霉素建立模型,10周末处死大鼠,无菌取肾脏,进行肾组织HE染色及masson染色,观察肾组织病理形态学改变,采用免疫组织化学及原位杂交方法分析肾组织内皮素-1蛋白及mRNA的表达。结果模型组大鼠肾组织肾小管上皮细胞(TEC)、肾小球系膜细胞(GMC)及肾小球上皮细胞(GEC)胞浆中ET-1蛋白及mRNA表达强阳性,三七总皂苷组大鼠肾组织ET-1蛋白及mRNA表达较模型组显著减少,有统计学意义(P<0.05)。结论三七总皂苷能够减少阿霉素肾病大鼠肾小管上皮细胞、肾小球系膜细胞及肾小球上皮细胞胞浆中内皮素-1蛋白及mRNA的表达,进而起到保护肾功能、延缓慢性肾衰竭的病程进展。
Objective To investigate the effects of panax notoginseng saponins on renal tissue endothelin-1 protein and mRNA expression in rats with adriamycin-induced nephropathy. Methods Forty-five rats were randomly divided into 5 groups: normal group, sham operation group, model group, benazepril group and panax notoginseng group. The right kidney of the rats was surgically removed and adriamycin was injected intraperitoneally to establish a model. The rats were sacrificed at the end of the 10th week. The kidneys were taken aseptically and HE staining and masson staining were performed. The pathological changes of the kidneys were observed. The expression of endothelin-1 protein and mRNA in renal tissues was analyzed by immunohistochemistry and in situ hybridization. Results The expression of ET-1 protein and mRNA in cytoplasm of renal tubular epithelial cells (TEC), glomerular mesangial cells (GMC) and glomerular epithelial cells (GEC) was strongly positive in the rat model group. Panax notoginseng total saponins The expression of ET-1 protein and mRNA in the kidney tissue of the rats was significantly reduced compared with the model group, with statistical significance (P<0.05). Conclusion Panax notoginseng saponins can reduce the expression of endothelin-1 protein and mRNA in renal tubular epithelial cells, glomerular mesangial cells, and glomerular epithelial cells in rats with adriamycin-induced nephropathy, thereby protecting renal function. Delay the progression of chronic renal failure.
