目的探讨三七总皂甙能否抑制急性坏死性胰腺炎大鼠肺 NF-κB的活性,减轻 ANP 时肺脏病理损害。方法 SD 大鼠30只。随机分为3组:即假手术模型组(SO 组),急性坏死性胰腺炎组(ANP 组),三七总皂甙预处理组(PNS),每组10只。SO 组和 ANP 组造模前1 h 腹腔注射0.9%生理盐水(0.1 ml/100 g),PNS 组造模前1h 腹腔注射50 mg/ml 三七总皂甙(0.1 ml/100 g)。5%牛磺胆酸钠大鼠胆胰管逆行注射制备大鼠急性坏死性胰腺炎模型。SO 组进腹后仅翻动胰腺和十二指肠后关腹,不注入牛磺胆酸钠。各组造模后于6 h 点处死大鼠,取肺组织测定湿/干重比和测其中 NF-κB活性。结果 ANP 组造模后6 h 肺组织 NF-κB阳性细胞数明显高于 SO 组(P<0.01),经 PNS 处理,NF-κB阳性细胞数和阳性单位较 ANP 组明显下降(P<0.05)。结论 NF-κB在大鼠急性坏死性胰腺炎肺组织中明显激活;三七总皂甙可以在体内抑制 NF-κB在肺组织中的激活,减轻各脏器的病理损害,可望用于治疗 ANP。
Objective To investigate whether panax notoginseng saponins can inhibit the activity of lung NF-κB in rats with acute necrotizing pancreatitis, and reduce the pathological damage of lung during ANP. Methods 30 SD rats. They were randomly divided into 3 groups: sham-operated model group (SO group), acute necrotizing pancreatitis group (ANP group), and Panax notoginseng saponin pretreatment group (PNS), 10 in each group. In the SO group and the ANP group, 0.9% physiological saline (0.1 ml/100 g) was intraperitoneally injected 1 hour before the model was established. In the PNS group, 50 mg/ml panax notoginseng saponin (0.1 ml/100 g) was intraperitoneally injected 1 hour before the model establishment. Rat model of acute necrotizing pancreatitis was prepared by retrograde injection of 5% sodium taurocholate into the biliary and pancreatic ducts. After the stomach of the SO group was paniculated, only the pancreas and duodenum were turned and the abdomen was closed. Sodium taurocholate was not injected. Rats were sacrificed at 6 h after model establishment, and lung tissues were taken for determination of wet/dry weight ratio and NF-κB activity was measured. Results The number of NF-κB positive cells in the lung tissue was significantly higher in the ANP group than in the SO group 6 h after modeling (P<0.01). After PNS treatment, the number of NF-κB positive cells and the positive cells were significantly decreased compared with the ANP group (P<0.05). . Conclusion NF-κB is obviously activated in rat lung tissue of acute necrotizing pancreatitis. Panax notoginseng can inhibit the activation of NF-κB in lung tissue and reduce the pathological damage of various organs. It is expected to be used to treat ANP. .
