目的探讨冷冻左心室前壁与结扎前降支两种方法诱导大鼠心肌梗死后制作慢性心力衰竭模型的对比研究。方法选择8~10周龄SD健康大鼠120只,随机分为冷冻组(50只),结扎组(50只)、假手术组(20只)。手术后4周行心电图、心功能检查,取心脏组织行TTC、HE染色并观察其死亡率。结果手术后4周冷冻组大鼠死亡率为32%低于结扎组48%。与假手术组比较,冷冻组、结扎组大鼠心电图显示为心肌梗死,LVEF、左心室短轴缩短率明显降低,左心室舒张末直径明显升高,差异有统计学意义(P<0.01),冷冻组和结扎组大鼠心肌切片、TTC染色可见100%梗死区;梗死区HE染色主要为瘢痕组织,假手术组未见心肌梗死。结论冷冻法诱导的心力衰竭模型在心功能、心肌梗死面积等方面等同于结扎法,且死亡率明显减低,冷冻左心室前壁诱导大鼠心肌梗死后慢性心力衰竭模型较结扎法更好。
Objective To investigate the comparative study of two models of freezing left ventricular anterior wall and ligation before the descending branch to induce chronic heart failure model after myocardial infarction in rats. Methods 120 healthy SD rats aged 8 ~ 10 weeks were randomly divided into two groups: the frozen group (50), the ligation group (50) and the sham operation group (20). Four weeks after surgery, ECG and cardiac function tests were performed. TTC and HE staining of heart tissue were performed to observe the mortality. Results The mortality rate of frozen group was 32% lower than that of ligation group 4 weeks after operation. Compared with the sham operation group, the electrocardiogram of the frozen group and the ligation group showed myocardial infarction. LVEF, shortening of left ventricular short axis and left ventricular end diastolic diameter were significantly higher (P <0.01) Frozen group and ligation group rat myocardial slices, TTC staining showed 100% infarct area; infarct HE staining was mainly scar tissue, sham operation group no myocardial infarction. Conclusion The model of heart failure induced by freezing is equivalent to ligation in terms of cardiac function and myocardial infarction area, and the mortality rate is obviously reduced. Chronic heart failure model induced by freezing left ventricular anterior wall is better than ligation in rats after myocardial infarction.
