[目的]探讨肾癌患者围手术期外周血CK19mRNA和CK20mRNA的变化。[方法]采用RT-PCR方法检测141例肾癌患者术前血液样本,分析CK19mRNA和CK20mRNA与临床、病理因素的关系;检测103例肾癌患者术前、术中和术后三个时间点的血液标本,并对三组标本进行比较。[结果]CK19mRNA、CK20mRNA的表达与患者的性别、年龄和肿瘤大小无关(P均>0.05);术后外周血CK19mRNA的拷贝数(5.68±3.08)高于术前(4.97±3.58),差异有统计学意义(P<0.05);肾癌患者外周血CK20mRNA的拷贝数低于CK19mRNA。[结论]检测肾癌患者循环肿瘤细胞CK19mRNA,可反映肿瘤的血源性播散,判断病情。
[Objective] To investigate the changes of CK19mRNA and CK20mRNA in peripheral blood of patients with renal cell carcinoma during perioperative period. [Methods] Preoperative blood samples of 141 patients with renal cell carcinoma were detected by RT-PCR. The relationship between CK19mRNA and CK20mRNA and the clinical and pathological factors was analyzed. The expressions of CK19mRNA and CK20mRNA in 103 patients with renal cell carcinoma were detected at three time points before, during and after operation Blood samples, and three groups of specimens were compared. [Results] The expression of CK19 mRNA and CK20 mRNA was not related to the gender, age and tumor size (all P> 0.05). The CK19 mRNA copy number (5.68 ± 3.08) in postoperative peripheral blood was significantly higher than that before operation (4.97 ± 3.58) Statistical significance (P <0.05); CK20mRNA copy number of peripheral blood in patients with renal cell carcinoma was lower than CK19mRNA. [Conclusion] Detection of CK19 mRNA in circulating tumor cells in renal cell carcinoma patients can reflect the blood-borne tumor spread and judge the condition.
