本文研究环胞苷及其分解产物的分离分析方法,并初步探讨其分解反应的动力学。建立了用细筛目阳离子树脂层析柱收集阿糖脲苷峰洗脱液,以及用阴离子树脂层析柱收集环胞苷和阿糖胞苷各峰洗脱液,并分别用紫外分光光度法进行含量测定的方法,回收率在95~105%,变异系数在2%左右。采用双柱层析较单柱层析节省分析时间。我们用此法分析了环胞苷在酸性水溶液中的分解产物,并采用紫外光谱、红外光谱及与标准品核对柱层析保留时间的方法确定了其主要分解产物是阿糖胞苷及阿糖脲苷。明确了环胞苷分解的主要过程是先开环,后脱氨的连续反应。
In this paper, the methods for the separation and analysis of cyclocytidine and its decomposition products are studied, and the kinetics of its decomposition reactions are preliminarily explored. A high-performance cation resin chromatography column was used to collect the arabinuridine peak eluent, and an anion resin column was used to collect the eluents of each of the cyclocytidine and cytarabine, respectively, and they were respectively subjected to ultraviolet spectrophotometry. The content determination method has a recovery rate of 95 to 105% and a coefficient of variation of about 2%. Using dual-column chromatography saves analysis time compared to single-column chromatography. We used this method to analyze the decomposed products of cyclocytidine in acidic aqueous solution, and determined the main decomposition products of cytarabine and arabinose using ultraviolet and infrared spectroscopy and the retention time of the column chromatography with the standard product.
