目的 探讨白细胞介素 1(IL 1)对骨髓瘤细胞粘附诱导骨髓基质细胞 (BMSCs)分泌白细胞介素 6(IL 6 )的影响及多发性骨髓瘤 (MM )患者和正常对照BMSCs分泌IL 6的差异。方法 建立人BMSCs培养方法 ,采用MTT比色法检测BMSCs培养上清中IL 6量。结果 (1)MM患者BMSCs具有异常增殖能力 ;(2 )单纯KM 3细胞培养上清中IL 6量为 7± 2 .3u/ml;(3)单纯BMSCs培养时 ,MM患者BMSCs分泌IL 6量为 2 6±7.5u/ml,高于正常对照 16± 4.7u/ml(P <0 .0 1) ;(4)KM3细胞与MM患者或正常对照BMSCs共培养 2 4小时后 ,其培养上清中IL 6量分别为 49± 18.1u/ml和 2 7± 6 .9u/ml,均高于单纯BMSCs培养上清中IL 6量 (P<0 .0 1) ;(5 )将BMSCs经 1%多聚甲醛固定 ,再与KM 3细胞共培养 2 4小时后 ,其培养上清中IL 6量与单纯KM3细胞培养上清比较差异无显著性 ;(6 )单纯BMSCs培养体系中加入IL 1,IL 6分泌量差异无显著性 ;(7)在KM 3细胞与BMSCs共培养体系中加入IL 1,IL 6分泌量达 74± 2 0 .5u/ml(MM组 )和 46± 10 .5u/ml(正常对照组 ) ,均高于单纯KM 3与BMSCs共培养 (P <0 .0 1)。结论 MM患者BMSCs具有异常增殖和分泌IL 6能力 ,IL 1通过骨髓瘤细胞介导可促进BMSCs分泌IL 6
