目的:探讨白英提取物(Solanum lyratum Thunb extract,STE)对人胃癌SGC-7901细胞凋亡及凋亡相关蛋白Survivin和Caspase-3表达的影响。方法:体外培养SGC-7901细胞,以2.5、5、10mg/L STE处理SGC-7901细胞48小时,并以2.5mg/L顺铂(DDP)作为阳性对照。用MTT法检测细胞抑制率,TUNEL法检测凋亡率,二步法免疫组化检测Survivin和Caspase-3表达。结果:与正常对照组相比,STE各浓度组细胞抑制率显著上升(P<0.001),细胞凋亡率明显升高(P<0.01),Caspase-3蛋白表达显著上升(P<0.05或P<0.01),Survivin蛋白表达明显下降(P<0.05或P<0.01)。结论:STE具有诱导SGC-7901细胞凋亡的作用,其分子机制可能与上调Caspase-3及下调Survivin蛋白表达有关。
Objective: To investigate the effect of Solanum lyratum Thunb extract (STE) on the apoptosis of human gastric cancer SGC-7901 cells and the expression of apoptosis-related proteins Survivin and Caspase-3. METHODS: SGC-7901 cells were cultured in vitro and SGC-7901 cells were treated with 2.5, 5, and 10 mg/L STE for 48 hours, and 2.5 mg/L cisplatin (DDP) was used as a positive control. The cell inhibition rate was detected by MTT assay, the apoptosis rate was detected by TUNEL method, and the expression of Survivin and Caspase-3 was detected by two-step immunohistochemistry. RESULTS: Compared with the normal control group, the inhibition rate of the STE concentration in each group significantly increased (P<0.001), the apoptosis rate increased significantly (P<0.01), and the expression of Caspase-3 protein increased significantly (P<0.05 or P). <0.01), Survivin protein expression was significantly decreased (P <0.05 or P <0.01). Conclusion: STE can induce apoptosis of SGC-7901 cells, and its molecular mechanism may be related to up-regulation of Caspase-3 and down-regulation of Survivin protein expression.
