许多肽类激素当以超生理浓度注入体内时都能调节它们在相应靶细胞上的受体。最近一些实验室报导,给完整雄性或雌性大鼠注射LHRH或其高活性类似物,导致睾丸及卵巢上促性腺激素的受体丧失,伴有性激素合成减少,副性器官萎缩等反应。本研究在未成年大鼠进一步观察了注射LHRH类似物对卵巢的LH/HCG受体,离体卵巢对LH/HCG的反应性以及cAMP积累和孕酮产生等方面的影响。作者给25天的雌性大鼠注射一种LHRH的高活性类似物(第10位去甘氨酸的LHRH乙基胺),或是单次注射1μg,或是每24小时重复注射1μg反复多次用药。对照动物注射0.1ml磷酸缓冲液。分别从实验及对照动物的卵巢制备出部分纯化细胞膜
Many peptide hormones, when injected into the body in super physiological concentrations, regulate their receptors on the corresponding target cells. Recently, some laboratories reported that injecting LHRH or its highly active analogues into intact male or female rats resulted in the loss of receptors for testosterone and ovarian gonadotropin, with decreased synthesis of sex hormones and atrophy of the accessory sex. In this study, the effects of LHRH analogue injection on ovarian LH / HCG receptor, ovarian response to LH / HCG, cAMP accumulation and progesterone production in juvenile rats were further observed. The authors injected a 25 day old female rat with a highly active analogue of LHRH (LHRH ethylamine at the 10th position to glycine), either as a single injection of 1μg, or as multiple injections of 1μg repeated every 24 hours. Control animals were injected with 0.1 ml phosphate buffer. Partially purified cell membranes were prepared from the ovaries of experimental and control animals, respectively
