目的:探讨二甲双胍联合卡铂对三阴性乳腺癌细胞MDA-MB-231增殖的影响。方法:体外培养三阴性乳腺癌细胞MDA-MB-231,分别给予不同浓度(0、2.5、5、10 mmol/L)二甲双胍和20μmol/L卡铂处理后,采用MTT实验、平板克隆实验检测二甲双胍联合卡铂对MDA-MB-231细胞增殖的影响。结果:MTT实验结果显示:二甲双胍抑制MDA-MB-231细胞的增殖,5、10mM组细胞OD490值均显著低于对照组(P<0.05),且10 mM组细胞OD490值明显低于其他各组(P<0.05);与单药相比,二甲双胍和卡铂联用组细胞生长抑制率显著升高,差异具有统计学意义(P<0.05)。平板克隆实验结果显示:与单药相比,二甲双胍和卡铂联用组细胞的克隆形成抑制率率显著升高,差异具有统计学意义(P<0.05)。结论:二甲双胍联合卡铂能够有效的抑制三阴性乳腺癌细胞MDA-MB-231的增殖。
Objective: To investigate the effect of metformin combined with carboplatin on the proliferation of triple negative breast cancer cell line MDA-MB-231. Methods: The MDA-MB-231 cells were cultured in vitro and treated with metformin at different concentrations (0,2.5,5,10 mmol / L) and carboplatin at 20μmol / L, MTT assay and plate clone assay were used to detect metformin Effect of carboplatin on the proliferation of MDA-MB-231 cells. Results: The results of MTT assay showed that metformin inhibited the proliferation of MDA-MB-231 cells and the OD490 values of 5 and 10mM groups were significantly lower than those of control group (P <0.05). The OD490 of 10mM group was significantly lower than that of other groups (P <0.05). Compared with single drug, the growth inhibition rate of metformin and carboplatin group was significantly increased, the difference was statistically significant (P <0.05). The results of plate cloning experiments showed that compared with single drug, the inhibitory rate of colony formation of metformin and carboplatin group was significantly increased, the difference was statistically significant (P <0.05). Conclusion: Metformin combined with carboplatin can effectively inhibit the proliferation of triple-negative breast cancer cells MDA-MB-231.
