1型糖尿病阴阳两虚证动物模型的制作与机理研究

目的:建立1型糖尿病阴阳两虚证动物模型,测定皮质醇、游离三碘甲状原氨酸(FT3)、胰岛素(INS)、血栓素B_2(TXB_2)、6-Keto-PGF_(1α)水平变化。方法:SD大鼠60只,链脲佐菌素(STZ)腹腔注射制作糖尿病模型(血糖≥16.7mmol/L)后,用中药青皮、枳壳、干姜、黄柏复方灌胃制作中医阴阳两虚证候模型,用金匮肾气丸进行反证治疗。结果:模型组、中药造模组大鼠血浆TXB_2、6-keto-PGF_(1α)及其比值较正常组均降低,但差异无显著性意义(P>0.05);金匮肾气丸组TXB_2值较模型组显著升高(P<0.05);6-Keto-PGF_(1α)值各组间比较,差异均无显著性意义(P>0.05)。模型组、中药造模组、金匮肾气丸组皮质醇较正常组显著增高(P<0.05);金匮肾气丸组较中药造模组有下降趋势。模型组FT3较正常组显著降低(P<0.05),金匮肾气丸组较正常组显著降低(P<0.05),较中药造模组有下降趋势。正常组、模型组、中药造模组胰岛素无显著改变,金匮肾气丸组显著升高(P<0.01)。电镜下可见模型组动物较正常组动物的胰岛、肾脏、肾上腺都发生了明显的病理改变。结论:中药青皮、枳壳、干姜、黄柏复方灌胃能制作糖尿病大鼠阴阳两虚证动物模型,其机理可能与皮质醇升高、FT3降低有关,金匮肾气丸有治疗作用,进一步说明该方法制作1型糖尿病阴阳两虚证候的动物模型的可行性。 OBJECTIVE: To establish an animal model of type 2 diabetes mellitus with deficiency of yin and yang to determine the change of cortisol, free triiodothyronine (FT3), insulin (INS), TXB_2, 6-Keto-PGF_ (1α) . Methods: Sixty Sprague Dawley rats were injected intraperitoneally with streptozotocin (STZ) to make a model of diabetes mellitus (blood glucose> = 16.7mmol / L). After the compound prescription of Qingpi, Fructus Aurantii, Syndrome model, anti-card with Jinkuishenqi pills. Results: The plasma levels of TXB_2, 6-keto-PGF_ (1α) and their ratios in model group and traditional Chinese medicine group were lower than those in normal group, but there was no significant difference (P> 0.05) The value of 6-Keto-PGF_ (1α) was significantly higher than that of the model group (P <0.05). There was no significant difference between the groups (P> 0.05). Compared with the normal group, the cortisol in the model group, the traditional Chinese medicine model group and the Jingui Shenqi Pill group were significantly higher than those in the normal group (P <0.05); the Jingui Shenqi Pills group showed a decreasing trend compared with the traditional Chinese medicine model group. Compared with the normal group, the FT3 in the model group was significantly lower than that in the normal group (P <0.05), while that in the Jingui Shenqi Pill group was significantly lower than that in the normal group (P <0.05). The normal group, the model group and the traditional Chinese medicine model group had no significant change of insulin, while the Jingui Shenqi Pill group increased significantly (P <0.01). Electron microscope shows that the model group of animals than the normal group of animals, islets, kidneys, adrenal gland have a significant pathological changes. Conclusion: The traditional Chinese medicine Qingpi, Fructus aurantii, Ginger, Treats can be made in rats with yin and yang deficiency syndrome animal model of diabetic rats, the mechanism may be related to increased cortisol, FT3 decreased, Jingui Shenqi Wan treatment, and further This method shows the feasibility of making animal model of type 1 diabetes mellitus with deficiency of yin and yang.
