以罗丹明B酰肼1和苯基乙二醛缩合反应得到席夫碱2,再经硼氢化钠还原得到新型罗丹明B衍生物R,采用~1H NMR、~(13)C NMR、ESI-MS、元素分析以及X-ray单晶射线对其结构进行了验证.研究结果表明,化合物R作为荧光探针能选择性地识别Hg~(2+),且受常见金属离子的干扰较小.探针R的荧光强度与Hg~(2+)浓度(5×10~(-7)~2.5×10~(-6)mol/L)呈良好的线性关系,并在MGC-803活细胞中实现了Hg~(2+)的成像.
Schiff base 2 was obtained by condensation reaction of rhodamine B hydrazide 1 with phenylglyoxal, and then a new rhodamine B derivative R was obtained by reduction with sodium borohydride. 1H NMR, 13 C NMR and ESI- MS, elemental analysis and single crystal X-ray diffraction.The results show that compound R can selectively recognize Hg 2+ as a fluorescent probe and has less interference with common metal ions. The fluorescence intensity of probe R showed a good linear relationship with the concentration of Hg 2+ (5 × 10 -7 ~ 2.5 × 10 -6 mol / L), and in the live cells of MGC-803 Hg ~ (2+) imaging was realized.
