目的探讨RNA干扰Th17关键转录因子ROR-γt表达后,对大鼠肾移植慢性排斥模型中Th17细胞功能以及慢性排斥反应的影响。方法构建ROR-γt特异性干扰腺病毒,将其通过尾静脉注入大鼠慢性排斥反应模型,监测大鼠血肌酐变化,酶联免疫吸附试验检测Th17相关细胞因子IL-17、IL-6和TNF-γ改变。4周后检测大鼠肾组织病理改变。结果成功构建ROR-γt特异性干扰腺病毒pSES-HUS-ROR-γt。大鼠慢性排斥反应模型尾静脉注入pSES-HUS-ROR-γt,大鼠血清肌酐水平下降,IL-17、IL-6和TNF-γ水平下降,肾小球纤维化病变减轻。结论 pSES-HUS-ROR-γt特异性干扰腺病毒可以抑制Th17细胞功能,减轻肾移植慢性排斥反应。
Objective To investigate the effect of R17-R17, a transcription factor of Th17, on Th17 cell function and chronic rejection in chronic rejection of renal allograft in rats. Methods ROR-γt-specific adenovirus was constructed and injected into the rat model of chronic rejection by tail vein to monitor the changes of serum creatinine. The levels of Th17-associated cytokines IL-17, IL-6 and TNF were measured by enzyme-linked immunosorbent assay -γ changes. After 4 weeks, pathological changes of rat kidney were detected. Results The ROR-γt-specific adenovirus pSES-HUS-ROR-γt was successfully constructed. Rats with chronic rejection model injected with pSES-HUS-ROR-γt through the tail vein, serum creatinine levels decreased, IL-17, IL-6 and TNF-γ levels decreased, glomerular fibrosis lesions reduced. Conclusion pSES-HUS-ROR-γt-specific adenovirus can inhibit the function of Th17 cells and reduce the chronic rejection of renal transplantation.
