Experimental studies have shown that leptin affects bone metabolism in mice and humans through a central mechanism. Leptin inhibits bone formation primarily by altering the central pathway of hypothalamic neuropeptide Y and alpha-MSH release; NPY and alpha-MSH have the opposite effect in regulating bone metabolism; leptin mediates body weight and bone weight Intermediate mediators are also regulators of energy deficits and neuroendocrine function, which regulate weight and bone mass via the central circuit.
