目的:探讨五味子多糖(Schisandra polysaccharide,SCP)对人结肠癌SW480细胞的增殖和凋亡的影响及其机制。方法:将人结肠癌SW480细胞分为对照组,SCP高剂量组、SCP中剂量组和SCP低剂量组,MTT法检测肿瘤细胞增殖情况,流式细胞仪检测凋亡,免疫组化法检测凋亡蛋白表达。结果:与对照组比较,SCP高、中、低剂量组均能不同程度抑制结肠癌SW480细胞的增殖,促进肿瘤细胞凋亡,使凋亡蛋白Bcl-2表达降低,Bax表达增加,具有剂量依赖关系。结论:五味子多糖能够抑制结肠癌SW480细胞的增殖同时促进细胞凋亡,这可能与其影响凋亡蛋白的表达相关。
Objective: To investigate the effects of Schisandra polysaccharide (SCP) on proliferation and apoptosis of human colon cancer SW480 cells and its mechanism. Methods: Human colon cancer SW480 cells were divided into control group, high dose SCP group, middle dose SCP group and low dose SCP group. The proliferation of SW480 cells was detected by MTT assay. Apoptosis was detected by flow cytometry and immunohistochemistry Death protein expression. Results: Compared with the control group, the high, middle and low doses of SCP could inhibit the proliferation of SW480 cells to a certain extent, promote the apoptosis of tumor cells, decrease the expression of Bcl-2 and increase the expression of Bax in a dose-dependent manner relationship. Conclusion: Schisandra polysaccharides can inhibit the proliferation of colon cancer SW480 cells and promote apoptosis, which may be related to its effect on the expression of apoptotic proteins.
