目的:探讨膀胱癌中长链非编码RNA(lnc RNA)TUC338的表达及作用机制。方法 :收集安徽医科大学一附院收治的30例膀胱癌手术患者的癌变组织和癌旁组织,通过实时定量PCR检测TUC338的表达水平并分析其临床病理特征之间的关联。运用RNA干扰(si-TUC338)的方法检测敲减TUC338,采用CCK8和半胱天冬酶3酶联免疫吸附测定法的方法分别检测TUC338对膀胱癌细胞的增殖、凋亡的影响。结果:相对于膀胱癌旁组织,TUC338 m RNA在膀胱癌组织中高表达。敲减TUC338后,细胞增殖被抑制,细胞凋亡增加。结论:TUC338是一个潜在的治疗膀胱癌的药物作用靶点。
Objective: To investigate the expression of long-chain noncoding RNA (TnnRNA) TUC338 in bladder cancer and its mechanism. Methods: Thirty cancerous tissues and adjacent tissues of bladder cancer patients were collected from the First Affiliated Hospital of Anhui Medical University. The expression of TUC338 was detected by real-time quantitative PCR and the correlation between the clinicopathological characteristics and the pathological changes was analyzed. TUC338 was knocked down by RNA interference (si-TUC338) assay. The effects of TUC338 on proliferation and apoptosis of bladder cancer cells were detected by CCK8 and caspase 3 enzyme-linked immunosorbent assay. Results: TUC338 m RNA was overexpressed in bladder cancer tissues compared with para-cancerous tissues. TUC338 knockdown, cell proliferation was inhibited, increased apoptosis. Conclusion: TUC338 is a potential therapeutic target for bladder cancer.
