目的了解SLE患者骨髓间质干细胞(MSC)的生物学特性。方法采用密度梯度离心和贴壁筛选法对9例SLE患者和5例正常人骨髓MSC进行分离培养,观察MSC形态,流式细胞术检测MSC表面分子CD29、CD44、CD34、CD45的表达及MTT法检测MSC的生长情况。结果 SLE患者MSC传代率(55.6%)低于正常(100%)(P<0.01),体外扩增成功的SLE患者MSC细胞形态、原代培养所需时间、平均传代时间及表面特异性分子表达均与健康对照组无明显差异。体外扩增不成功的SLE患者MSC易于自行凋亡。结论 SLE患者MSC有异质性,部分患者MSC存在生物学特性异常。
Objective To understand the biological characteristics of bone marrow mesenchymal stem cells (MSCs) in patients with SLE. Methods The MSCs of 9 patients with SLE and 5 normal controls were isolated and cultured by density gradient centrifugation and adherence screening. MSCs morphology was observed. The expression of CD29, CD44, CD34 and CD45 on MSC surface was detected by flow cytometry and MTT assay Detect the growth of MSC. Results The passage rate of MSCs in SLE patients (55.6%) was lower than that in normal subjects (100%) (P <0.01). The morphology of MSC cells, the time of primary culture, the average passage time and the surface-specific molecular expression No significant difference with the healthy control group. In vitro SLE patients with unsuccessful expansion of the MSCs are prone to apoptosis on their own. Conclusion MS patients with SLE have heterogeneity, some patients with abnormal biological characteristics of MSC.
