目的研究动态条件下Ⅱ级生物安全柜操作台面不同部位细菌污染情况。方法采用平板自然沉降法进行监测,设无实验者操作动态组、有实验者操作动态组,选取生物安全柜中5个监测点,在开始测试后30、60 min时间段进行空气沉降菌监测。结果在生物安全柜无实验操作的动态运行条件下,靠近操作面区域的菌落总数随时间推移而增加,靠近中部和内角区域的菌落总数随时间推移无显著增加。在生物安全柜有实验员进行正常操作的动态运行条件下,靠近操作面区域和左侧内角污染物存放区域的菌落总数随时间推移而增加,靠近中部和内角洁净物品存放区域的菌落总数随时间推移无显著增加。结论为避免Ⅱ级生物安全柜中的环境污染样品,要尽可能将实验操作部位设置在靠近生物安全柜内的中心区域。
Objective To study the bacterial contamination in different parts of the workbench of Class Ⅱ biological safety cabinet under dynamic conditions. Methods The natural plate sedimentation method was used for monitoring. Non-experimenter operation dynamic group was set up. Some experimenters operated dynamic group. Five monitoring points in biological safety cabinet were selected. Airborne sedimentation bacteria were monitored at 30 and 60 min after the start of test. Results The total number of colonies close to the operating area increased with the passage of time under the dynamic operating conditions of the biological safety cabinet without experimental operation. The total number of colonies near the middle and inner corner areas did not increase significantly with the passage of time. The total number of colonies near the operating area and the left corner of the contaminant storage area increased over time under dynamic operating conditions where the biological safety cabinet was under normal operating conditions of the experimenter. The total number of colonies near the central and inner corner of the clean item storage area increased with time No significant increase over time. Conclusion In order to avoid the environmental pollution in class Ⅱ biological safety cabinet samples, as far as possible the experimental operation site is set near the biological safety cabinet in the central area.
