随着基因工程技术的发展,蛋白质、多肽类药物的临床应用越来越受到重视。但蛋白质、多肽类药物的免疫原性和毒副反应及半衰期短等问题限制了其应用和发展。聚乙二醇(Polyethylene glycol,PEG)能够使蛋白药物溶解行为改善,稳定性增强,免疫原性消除或降低,循环半衰期延长,药代动力学优化,在生物技术和生物医学中具有广泛的应用前景。本文就蛋白质、多肽类药物的PEG修饰、修饰技术的发展、修饰的主要途径以及PEG修饰药物的现状及前景作一综述。
