目的观察气道平滑肌细胞(ASMC)经致敏血清刺激后嗜酸性粒细胞趋化因子(eotaxin)的表达变化及沙丁胺醇对其表达的影响,探讨其可能的作用机制及β2受体激动剂在哮喘治疗中的作用。方法无菌条件下分离SD大鼠气管段,组织块贴壁法体外培养ASMC。建立SD大鼠哮喘模型,采血收集血清。以致敏动物血清刺激体外培养的ASMC并收集培养上清及细胞。采用酶联免疫吸附法测定培养液上清中的eotaxin水平;放射免疫法测定培养细胞胞内环磷酸腺苷(cAMP)的表达;逆转录-聚合酶链反应半定量检测培养细胞中的eotaxin mRNA的表达。结果哮喘大鼠被动致敏后,气道平滑肌细胞可表达eotaxin(n=4,P<0.05),其表达水平与胞内的cAMP水平呈负相关(P<0.01,r=0.788);沙丁胺醇干预后,ASMC中eotaxin的表达明显下降(n=4,P<0.05)。结论气道平滑肌细胞在致敏状态下eotaxin的表达显著增强,作为炎性分泌细胞参与哮喘发生过程,是哮喘发病机制的重要组成部分。沙丁胺醇可以在哮喘发病中有效抑制炎性细胞因子的表达。
Objective To investigate the expression of eotaxin in asthmatic airway smooth muscle cells (ASMCs) stimulated by sensitized serum and the effect of salbutamol on the expression of eotaxin, and to explore its possible mechanism and the role of β2 agonists in asthma The role of treatment. Methods SD rat trachea was isolated under aseptic conditions and ASMC was cultured in vitro by tissue explant method. Establish SD rat asthma model, blood collection serum. The sensitized animal serum stimulated ASMC cultured in vitro and the culture supernatant and cells were collected. The levels of eotaxin in culture supernatant were determined by enzyme-linked immunosorbent assay (ELISA). The expression of cAMP in cultured cells was detected by radioimmunoassay. The expression of eotaxin mRNA in cultured cells was detected by reverse transcription-polymerase chain reaction expression. Results After sensitized asthmatic rats, airway smooth muscle cells could express eotaxin (n = 4, P <0.05), and the expression of eotaxin was negatively correlated with intracellular cAMP level (P <0.01, r = 0.788) , The expression of eotaxin in ASMC was significantly decreased (n = 4, P <0.05). Conclusion The expression of eotaxin in sensitized airway smooth muscle cells is significantly increased. As an inflammatory cytokine involved in the development of asthma, it is an important part of asthma pathogenesis. Salbutamol can effectively suppress the expression of inflammatory cytokines in the pathogenesis of asthma.
