目的:明确黄连超高效液相色谱-质谱联用技术(UPLC-MS/MS)指纹图谱与其抗炎药效的相关性,为黄连质量控制和药效物质基础研究提供科学依据。方法:采用灰色关联度分析法研究黄连UPLC-MS/MS指纹图谱和黄连对湿热型溃疡性结肠炎大鼠模型血清白细胞介素4(IL-4)影响的谱效相关性。结果:建立了黄连UPLCMS/MS指纹图谱的共有模式,确定了能够表征黄连特征的13个共有峰。与模型组比较,不同批次的黄连给药组均能不同程度的升高湿热型溃疡性结肠炎大鼠模型血清IL-4的含量(P<0.05,P<0.01)。黄连UPLC-MS/MS指纹图谱的各共有峰与其抗炎药效作用的关联度均>0.6。结论:通过谱效关系研究明确了黄连发挥抗炎药效作用是盐酸小檗碱、盐酸药根碱、黄连碱等多成分共同作用的结果。
OBJECTIVE: To clarify the correlation between the fingerprint of UPLC-MS / MS and anti-inflammatory effect of Coptis Chinensis Franch., And to provide a scientific basis for the research on the quality control and pharmacodynamics of Coptis chinensis. Methods: The gray relational analysis was used to study the relationship between the chromatogram of UPLC-MS / MS and the effect of Coptis chinensis Franch on serum interleukin 4 (IL-4) in rat models of damp-heat ulcerative colitis. Results: The consensus model of UPLCMS / MS fingerprinting was established and 13 common peaks were identified. Compared with the model group, different batches of Coptis administration group could all increase the level of IL-4 in the rat model of damp-heat ulcerative colitis (P <0.05, P <0.01). The correlations between the common peaks of Coptis chinensis UPLC-MS / MS fingerprints and their anti-inflammatory pharmacological effects were both> 0.6. Conclusion: It is clear from the spectrum-effect relationship that the anti-inflammatory effect of Coptis chinensis is the result of the combined action of berberine hydrochloride, jatrorrhizine hydrochloride and coptisine.
