目的运用Identifiler和STRtyper-10F/G扩增试剂盒用于单亲亲子鉴定500例,探索两试剂盒联用在亲子鉴定案例中的应用价值。方法提取血样本基因组DNA,采用荧光标记STR-PCR复合扩增、毛细管电泳基因分型技术,应用ABI3130遗传分析仪对500例单亲亲子鉴定的个体进行24个STR基因座的基因分型。结果在500例单亲亲子鉴定中,否定亲权的80个案例中均至少有5个基因座不符;认定亲权的420例中,累积父权指数均>10000。在认定的亲子关系中,有17例出现1个基因座的基因突变,其中一步突变16例,两步突变1例。结论联合应用Identifiler和STRtyper-10F/G体系的24个STR基因座的基因鉴定,能成功地开展单亲亲子鉴定。
Objective To identify 500 single-parent paternity tests by using Identifiler and STRtyper-10F / G amplification kit and to explore the value of the two kits in the paternity test. Methods Genomic DNA was extracted from blood samples and genotyped 24 STR loci in 500 individuals with single parentage by ABI3130 genetic analyzer using fluorescence-labeled STR-PCR multiplex amplification and capillary electrophoresis genotyping. Results In the 500 cases of single-parent paternity test, at least 5 loci were out of 80 cases of negative parental authority. Among the 420 cases of parental authorization, the cumulative paternity index was> 10000. In the identified paternity, there are 17 cases of gene mutation at a locus, including one step mutation in 16 cases, two mutations in 1 case. Conclusion The genetic identification of 24 STR loci using Identifiler and STRtyper-10F / G system can successfully carry out single-parent paternity test.
