目的:观察1,25(OH)2VitD3、顺铂对人卵巢癌细胞株增殖及凋亡的作用及Ki-67,Bax蛋白表达情况。方法:MTT法检测1,25(OH)2VitD3单独及其与顺铂合用后卵巢癌细胞的抑制率、流式细胞术测定细胞周期及凋亡率、免疫组化法检测Ki-67,Bax蛋白表达情况。结果:1,25(OH)2VitD3、顺铂均可抑制HO-8910细胞的生长(P<0.01),阻滞细胞于G0/G1期并诱导HO-8910细胞凋亡(P<0.01),当两者合用时Ki-67几乎不表达,而Bax蛋白表达增强(P<0.01)。结论:1,25(OH)2VitD3与顺铂合用有协同抑制增殖和诱导凋亡的作用。
Objective: To investigate the effects of 1,25 (OH) 2VitD3 and cisplatin on the proliferation and apoptosis of human ovarian cancer cell lines and the expression of Ki-67 and Bax protein. Methods: The inhibitory rates of 1,25 (OH) 2VitD3 alone and in combination with cisplatin on the proliferation of ovarian cancer cells were determined by MTT assay. The cell cycle and apoptosis rate were determined by flow cytometry. The expressions of Ki-67 and Bax protein were detected by immunohistochemistry Express the situation. Results: 1,25 (OH) 2VitD3 and cisplatin both inhibited the growth of HO-8910 cells (P <0.01), blocked the cells in G0 / G1 phase and induced the apoptosis of HO-8910 cells (P <0.01) When combined, Ki-67 was almost not expressed, while Bax protein expression was enhanced (P <0.01). Conclusion: The combination of 1,25 (OH) 2VitD3 and cisplatin can inhibit proliferation and induce apoptosis.
