目的研究外源激素、水杨酸、茉莉酸甲酯对海巴戟天悬浮细胞生长及蒽醌类化合物合成的影响。方法建立海巴戟天细胞悬浮培养体系,并向海巴戟天细胞悬浮培养物中添加外源激素、水杨酸和茉莉酸甲酯,测定细胞比生长速率和蒽醌产量。结果外源激素萘乙酸(NAA)、激动素(KT)对海巴戟天悬浮细胞的生长和蒽醌类化合物的合成有显著影响,采用B5培养基时,较佳激素组合为NAA 2 mg/L+KT 0.1 mg/L。在对数生长初期(培养9 d)时,添加200μmol/L水杨酸和50μmol/L茉莉酸甲酯,蒽醌质量浓度分别为588.34 mg/L和595.49 mg/L,比对照分别提高了17.05%和18.47%。结论适宜激素配比结合一定质量浓度的水杨酸和茉莉酸甲酯外源诱导对蒽醌类化合物的合成有促进作用。
Objective To study the effects of exogenous hormones, salicylic acid and methyl jasmonate on the growth of suspension cells and synthesis of anthraquinones in Morinda officinalis. Methods To establish the suspension culture system of Morinda citrifolia. The exogenous hormones, salicylic acid and methyl jasmonate were added to the suspension cultures of Morinda citrifolia. The specific growth rate and anthraquinone yield were determined. Results NAA and KT had significant effects on the growth of suspension cells and the synthesis of anthraquinones in the suspension of Haemaria morindae. The best combination of hormones was NAA 2 mg / L + KT 0.1 mg / L. At the beginning of logarithmic growth (cultured for 9 d), the concentration of anthraquinones were 588.34 mg / L and 595.49 mg / L, respectively, when 200 μmol / L salicylic acid and 50 μmol / L methyl jasmonate were added, % And 18.47%. Conclusion The combination of appropriate hormones with a certain concentration of salicylic acid and methyl jasmonate induce the synthesis of anthraquinones.
