高血压是常见病,急需寻找新的降压药。在寻找新药的筛选中,急性实验动物静脉注射试药,对不纯的、作用慢的、不完全溶解的药很不相宜;慢性实验性高血压动物试药,工作量太大。权衡利弊,根据等毒性剂量比较疗效的原则,提出大白鼠多次灌胃给药、一次直接测量血压的方案,作为降压药的初筛方法。实验设计先作小鼠毒性试验,灌胃给药1次,算出半数致死量。取其1/5的剂量给大鼠灌胃3天(例如小鼠半数致死量为0.5克/公斤,则大鼠每天灌胃0.1克/公斤),麻醉后直接测量动脉血压。
