本文用戊二醛两步法与改良过碘酸氧化法制备兔抗小鼠IgG与辣根过氧化物酶(HRP)的结合物,再用SDS-聚丙烯酰胺凝胶电泳与酶联免疫电转技术检测了结合物的均一性。结果表 明。戊二醛两步法制备的结合物产率较低,活性不高;而改良过碘酸氧化法得到的结合物产率高,抗体与酶的活性保持稳定。本文还对两种方法的机制进行了探讨。
In this paper, the conjugate of rabbit anti-mouse IgG and horseradish peroxidase (HRP) was prepared by a two-step glutaraldehyde method and a modified periodate oxidation method. Then SDS- polyacrylamide gel electrophoresis and enzyme-linked immunosorbent assay The technique detects the homogeneity of the conjugate. The results show. Glutaraldehyde two-step preparation of conjugates with low yield, activity is not high; and improved periodic acid oxidation obtained conjugate yield, antibody and enzyme activity remained stable. The paper also discusses the mechanism of the two methods.
