目的观察二甲双胍(MF)对去势大鼠血清中TNF-α,IL-6及瘦素含量的影响,探讨MF对去势大鼠瘦素抵抗的改善作用和免疫学机制。方法选取Wistar雌性去势大鼠30只,随机分为去势模型组、MF低剂量组(MFL)和MF高剂量组(MFH),每组10只,另选健康Wistar雌性大鼠10只为假手术对照组,用药组分别给予MF 135mg·kg~(-1)·d~(-1),270mg·kg~(-1)·d~(-1),去势模型组和假手术对照组分别给予等容量生理盐水,灌胃,连续给药42d。采用ELISA法,测定大鼠血清TNF-α及IL-6含量;采用RIA法测定大鼠血清瘦素水平。结果与假手术对照组比较,去势大鼠血清TNF-α,IL-6及瘦素含量显著升高,同时体重明显增加(P<0.05);与去势组比较,二甲双胍高剂量组血清TNF-α,IL-6及瘦素水平则明显降低,体重亦显著降低(P<0.05),而二甲双胍低剂量组无显著变化(P>0.05)。结论 MF可显著降低去势大鼠血清瘦素含量,降低去势大鼠体重,其机制与调节去势大鼠血清TNF-α及IL-6水平有关。
Objective To observe the effects of metformin (MF) on the levels of serum TNF-α, IL-6 and leptin in ovariectomized rats and the possible mechanism of leptin resistance. Methods Thirty Wistar female castrated rats were randomly divided into castrated model group, MF low-dose group (MFL) and MF high-dose group (MFH), 10 rats in each group, and 10 healthy Wistar female rats The sham operation control group and the medication group were given MF 135 mg · kg -1 · d -1 and 270 mg · kg -1 · d -1 respectively. The model group and the sham operation control group Group were given equal volume of saline, gavage, continuous administration 42d. Serum TNF-α and IL-6 levels were measured by ELISA. Serum leptin levels were measured by RIA. Results Compared with sham control group, the levels of serum TNF-α, IL-6 and leptin in ovariectomized rats increased significantly and the body weight increased significantly (P <0.05). Compared with sham-operated group, α, IL-6 and leptin levels were significantly lower, body weight was significantly lower (P <0.05), and metformin low dose group had no significant change (P> 0.05). Conclusion MF can significantly reduce the serum leptin content in ovariectomized rats and reduce body weight in ovariectomized rats. The mechanism may be related to the regulation of serum TNF-α and IL-6 levels in ovariectomized rats.
