目的 探讨卡托普利 (CPT)对野百合碱 (MCT)诱发大鼠慢性炎性肺动脉高压的影响。方法 将33只雄性Wister大鼠随机分为正常对照组、肺动脉高压模型组、CPT治疗组 ,每组 11只。予MCT(5 0mg/kg)复制大鼠肺动脉高压模型后 ,分别给CPT组及模型组大鼠连续灌胃CPT 2 5mg/ (kg·d)或等量生理盐水2 1d ,采用改良右心导管术测定肺血流动力学参数 ,以非平衡法测定静脉血浆内皮素 - 1(ET - 1)水平 ;处死大鼠后 ,称量肺湿重 (wW )、右心室自由壁 (RV)和左心室加室间隔 (LV +S)重 ,计算右心肥厚指数 (RV/LV +S)。结果 CPT能明显降低肺动脉高压模型大鼠的平均肺动脉压、wW、RV/LV +S及血浆ET - 1水平 (P <0 .0 5 ,或P <0 .0 1)。结论 长期使用CPT能有效降低MCT所致肺动脉高压 ,改善心功能 ,其作用机制可能与显著降低血浆ET - 1水平有关。
Objective To investigate the effect of captopril (CPT) on monocrotaline (MCT) -induced chronic inflammatory pulmonary hypertension in rats. Methods Thirty-three male Wister rats were randomly divided into normal control group, pulmonary hypertension model group and CPT treatment group, 11 rats in each group. After MCT (50 mg / kg) was given to rat model of pulmonary hypertension, CPT group and model group were given continuous CPT 25 mg / (kg · d) or normal saline for 21 days respectively. The modified right heart catheter The parameters of pulmonary hemodynamics were measured and the level of endothelin - 1 (ET - 1) in venous plasma was measured by non - equilibrium method. After the rats were sacrificed, the wet weight of lung (wW), right ventricular free wall (RV) Ventricular plus interventricular septum (LV + S) weight, calculate right ventricular hypertrophy index (RV / LV + S). Results CPT significantly reduced mean pulmonary arterial pressure, wW, RV / LV + S and plasma ET - 1 levels (P <0.05 or P <0.01) in pulmonary hypertensive rats. Conclusion Long - term use of CPT can effectively reduce pulmonary hypertension induced by MCT and improve cardiac function, which may be related to the significant reduction of plasma ET - 1 level.
