[目的]探讨顺铂诱导卵巢癌细胞凋亡作用的机制。[方法]应用不同浓度顺铂处理卵巢癌HO鄄8910细胞,于不同时间收集细胞,分别进行倒置显微镜观察活细胞,Gimsa染色观察细胞形态改变并计数凋亡率,琼脂糖凝胶电泳观察DNA降解情况,SP免疫组化染色观察凋亡过程中p53蛋白的表达情况。部分涂片还使用了TUNEL染色计数凋亡细胞,并与Gimsa染色计数的凋亡细胞数作比较。[结果]在顺铂作用下,卵巢癌HO鄄8910细胞呈现典型的凋亡细胞形态学改变,如细胞核固缩、染色体凝聚、有凋亡小体形成,细胞DNA电泳呈梯形带,凋亡率及凋亡相关蛋白p53表达阳性率随着药物作用时间延长及药物浓度增大而升高,且二者呈正相关。TUNEL染色计数的凋亡细胞个数明显高于Gimsa染色。[结论]顺铂可以通过诱导卵巢癌细胞系HO鄄8910细胞凋亡而发挥其抗肿瘤功效,且为p53依赖性细胞凋亡过程。
[Objective] To investigate the mechanism of cisplatin-induced ovarian cancer cell apoptosis. [Methods] HO-8910 cells were treated with different concentrations of cisplatin. Cells were collected at different time points. The cells were observed under inverted microscope. The morphological changes were observed by Gimsa staining and the apoptosis rate was counted. The DNA degradation was observed by agarose gel electrophoresis SP immunohistochemistry was used to observe the expression of p53 protein during apoptosis. Some smears also used TUNEL stain to count apoptotic cells, and compared with the number of apoptotic cells counted by Gimsa staining. [Results] Under the action of cisplatin, ovarian cancer HO-8910 cells showed morphological changes of typical apoptotic cells, such as nuclear pyknosis, chromosome condensation, formation of apoptotic bodies, cell DNA electrophoresis with trapezoidal band, apoptosis rate And the positive rate of apoptosis related protein p53 expression increased with the prolongation of drug effect and drug concentration, and the positive correlation between the two. The number of apoptotic cells by TUNEL staining was significantly higher than Gimsa staining. [Conclusion] Cisplatin can exert its anti-tumor effect by inducing apoptosis of ovarian cancer cell line HO-8910, and is a p53-dependent apoptotic process.
