目的:检测转化生长因子-β1(transforming growth factor-beta 1,TGF-β1)在猪冠状动脉微栓塞模型中的表达情况。方法:10只小型猪,通过经皮冠脉介入术于前降支注入微栓塞球建立冠状动脉微栓塞的模型,以ELISA法检测术前、术后2h、6h和1周时血清TGF-β1的浓度;RT-PCR法检测术后1周时心肌中TGF-β1 mRNA的表达情况。结果:与冠脉微栓塞术前(7.38±2.10ng·mL-1)相比,在术后2h血清中TGF-β1升高(14.86±4.04ng·mL-1),6h达到高峰(22.89±2.03ng·mL-1),1周时下降(20.44±3.99ng·mL-1),但仍高于术前;RT-PCR法检测示1周时前壁微梗死区心肌组织TGF-β1 mRNA水平表达高于后壁未梗死区。结论:冠状动脉微栓塞后血清TGF-β1呈升高趋势,且至少持续1周左右;1周时微栓塞相关心肌组织中TGF-β1 mRNA的表达也明显升高。
Objective: To investigate the expression of transforming growth factor-beta 1 (TGF-β1) in porcine coronary micro-embolism model. Methods: 10 miniature pigs were established coronary micro-embolization model by percutaneous coronary intervention on the anterior descending branch with micro-embolism ball. Serum TGF-β1 levels were measured by ELISA before, 2h, 6h and 1 week The mRNA expression of TGF-β1 in myocardium was detected by RT-PCR at 1 week after operation. Results: TGF-β1 increased (14.86 ± 4.04 ng · mL-1) at 2 h after operation and peaked at 6 h (22.89 ± 2.03ng · mL-1), and decreased at 1 week (20.44 ± 3.99ng · mL-1), but still higher than that before operation. The expression of TGF-β1 mRNA in myocardial infarction The level of expression is higher than the posterior wall without infarction area. CONCLUSION: The serum TGF-β1 level in coronary micro-embolism is increasing at least for about 1 week. The expression of TGF-β1 mRNA in myocardial tissue associated with microembolization is also significantly increased at 1 week.
