目的研究南蛇藤总萜提取物对肝癌7721细胞侵袭和黏附能力的影响。方法南蛇藤总萜提取物作用肝癌7721细胞后,MTT法及细胞黏附法检测细胞毒作用及对黏附能力的影响;流式细胞仪检测基质金属蛋白酶-2(MMP-2)表达的变化;ELISA法检测血管内皮生长因子(VEGF)分泌量的变化。结果10、20、40、80μg/mL南蛇藤总萜提取物作用肝癌7721细胞后,其生长抑制率分别为(8.62±0.87)%、(13.42±1.07)%、(27.64±2.13)%(、47.16±3.08)%,呈剂量依赖性。南蛇藤总萜提取物可以抑制肝癌7721细胞与人脐静脉内皮细胞和基底膜成分Matrigel的侵袭、黏附能力,能明显下调MMP-2和VEGF的表达,且呈一定的量效关系。结论南蛇藤总萜提取物可以抑制肝癌7721细胞增殖,降低细胞的侵袭、黏附能力,其机制可能与下调VEGF和MMP-2的表达有关。
Objective To study the effects of extracts from the total scorpion of Nan snake vine on the invasion and adhesion of hepatoma 7721 cells. Methods The effects of the total extracts of Nancao rattan on liver cancer 7721 cells were detected by MTT assay and cell adhesion assay. The changes of matrix metalloproteinase-2 (MMP-2) expression were detected by flow cytometry. The change of vascular endothelial growth factor (VEGF) secretion was detected by ELISA. Results After 10, 20, 40, and 80 μg/mL extracts of total snakeheads of Nancao rata, the growth inhibition rates of liver cancer 7721 cells were (8.62±0.87)%, (13.42±1.07)%, and (27.64±2.13)%, respectively. 47.16±3.08)% in a dose-dependent manner. The extract of total snake radix sinensis could inhibit the invasion and adhesion ability of hepatoma 7721 cells and human umbilical vein endothelial cells and Matrigel, and significantly down-regulate the expression of MMP-2 and VEGF, and showed a dose-effect relationship. Conclusion The extract of total snake vines of Nan snake vine can inhibit the proliferation of hepatocellular carcinoma 7721 cells and reduce the invasion and adhesion of cells. The mechanism may be related to the down-regulation of VEGF and MMP-2 expression.
