目的:针对人结肠癌HCT-116细胞在体外培养时常出现部分细胞聚团生长的特性,建立一种简易的细胞传代培养方法———水平震荡法。方法:采用简单高效的水平震荡方法替代胰酶消化法对HCT-116细胞进行传代培养。结果:利用水平震荡方法传代后的细胞在新瓶中可以正常贴壁。24h和48h观察到胰酶消化法传代的细胞生长情况与水平震荡法传代的细胞未见明显差异。结论:该方法无需使用胰蛋白酶,避免了胰酶对细胞的损伤作用,而且操作简便,降低了实验成本和污染的风险。水平震荡传代法可以替代胰酶消化法作为HCT-116细胞的简易传代方法。
OBJECTIVE: To establish a simple cell subculture method --- horizontal shock method based on the characteristics of human colon cancer HCT-116 cells cultured in vitro. Methods: HCT-116 cells were subcultured by simple and efficient horizontal shaking instead of trypsin digestion. Results: The passaged cells using the horizontal shaking method can adhere normally in the new bottle. 24h and 48h observed pancreatic enzyme digestion passage cell growth and horizontal shock method passage of cells did not show significant differences. Conclusion: This method does not require the use of trypsin, pancreatic enzymes to avoid damage to cells, but also easy to operate, reducing experimental costs and the risk of contamination. Horizontal oscillatory passaging can replace pancreatin digestion as a simple passaging method for HCT-116 cells.
