初步探讨重组生长激素 (rGH )改善肝切除术后疲劳综合征 (POF )的分子机制及对肝脏再生的影响。方法 采用半定量逆转录聚合酶链式反应 (RT PCR )技术 ,分别对治疗组及对照组大鼠切肝 4 0 %术后 3 ,7d生长激素受体mRNA (GHRmRNA )的表达及相对肝重量 (肝重 体重 )进行研究。结果 手术后 3d组GHRmRNA表达量明显下降 ;治疗组术后3dGHRmRNA的表达和术后 7d肝脏再生较对照组明显增加 (P <0 .0 5)。结论 术后GHRmRNA的低水平表达 ,可能是导致POF发生的机制之一 ;rGH的应用能刺激术后大鼠肝脏GHRmRNA的表达及促进肝脏的再生
To investigate the molecular mechanism of recombinant growth hormone (rGH) improving post-hepatectomy-induced fatigue syndrome (POF) and its effect on liver regeneration. Methods Semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) was used to detect the expression of growth hormone receptor mRNA (GHR mRNA) and the relative weight of liver in the treated group and the control group at 40% (Liver weight) to study. Results The expression of GHR mRNA decreased significantly in the 3d group after operation. The expression of 3dGH mRNA and the hepatic regeneration 7 days after operation in the treatment group were significantly higher than those in the control group (P <0.05). Conclusions The low level expression of GHR mRNA may be one of the mechanisms leading to POF. The application of rGH can stimulate the expression of GHR mRNA and promote the regeneration of liver in postoperative rats
