目的:研究利奈唑胺(LZD)对严重烧伤家兔促炎症因子肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白介素-6(IL-6)水平的影响。方法:随机将24只家兔分为假烫组、烫伤组、假烫+LZD治疗组、烫伤+LZD治疗组4组,每组6只。将两烫伤组家兔背部皮肤置于98℃热水18 s制备30%TBSAⅢ度烫伤模型;假烫组用37℃温水代替热水。两LZD治疗组家兔静脉注射10 mg/kg利奈唑胺。组织病理学观察烫伤深度,采用放射免疫分析方法检测烧伤后家兔血清TNF-α、IL-1、β、IL-6水平。结果:与假烫组家兔比较,烧伤家兔TNF-α、IL-1β及IL-6显著升高,IL-1β在伤后12 h达峰值,TNF-α及IL-6在伤后24 h达峰值。利奈唑胺能显著降低烫伤家兔TNF-α、IL-1β及IL-6水平(P<0.05),降幅分别为28.19%、38.54%和28.48%,但利奈唑胺对假烫家兔3种细胞因子的水平无显著影响(P>0.05)。结论:利奈唑胺能显著降低烫伤家兔的促炎症因子水平。
Objective: To investigate the effect of linezolid (LZD) on the levels of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and interleukin-6 (IL-6) in rabbits with severe burns. Methods: Twenty-four rabbits were randomly divided into four groups: fake-hot group, scald group, fake-hot + LZD group and burn + LZD group. The scalded skin of rabbits in two scalded groups were placed in 98 ℃ hot water for 18 s to prepare 30% TBSA Ⅲ degree scald models; fake hot group was replaced by hot water at 37 ℃. Two LZD treatment rabbits intravenously with 10 mg / kg linezolid. Histopathology was used to observe the scald depth. Serum levels of TNF-α, IL-1, β and IL-6 were detected by radioimmunoassay. Results: The levels of TNF-α, IL-1β and IL-6 in burned rabbits were significantly increased compared with those in sham-burn group, the levels of IL-1β peaked at 12 hours and the levels of TNF-α and IL- h up to the peak. Linezolid significantly reduced levels of TNF-α, IL-1β and IL-6 in burned rabbits (28.19%, 38.54% and 28.48%, respectively) Cytokines levels had no significant effect (P> 0.05). Linezolid can significantly reduce the levels of pro-inflammatory cytokines in burned rabbits.
