目的:观察中药不同配伍方式对糖尿病大鼠的影响,探讨影响七箭方药效的因素。方法:高脂饲料加小剂量链脲佐菌素注射法建立糖尿病大鼠模型,以改良的均匀设计法设计药物的5种不同组合方式,不同组合药物灌服30d,检测血糖、甘油三酯、胆固醇、血肌酐、尿肌酐、尿白蛋白,计算肌酐清除率,对实验结果进行回归分析。结果:试验组1-5比模型组血糖、甘油三酯、胆固醇、尿微量白蛋白明显降低,有统计学意义(P<0.05或P<0.01);试验组3-5比模型组肌酐清除率升高,有统计学意义(P<0.05或P<0.01)。根据回归模型分析对不同指标的影响因素,结果黄连小檗碱对血糖的影响最大;对甘油三酯的重要影响因素依次为天花粉多糖、黄连小檗碱、鬼箭羽黄酮;对胆固醇的重要影响因素依次为黄连小檗碱、天花粉多糖、鬼箭羽黄酮;对肌酐清除率的重要影响因素为三七皂苷;对尿微量白蛋白的重要影响因素依次为鬼箭羽黄酮、黄连小檗碱、三七皂苷、天花粉多糖。结论:改良的均匀设计法简便可行,对糖尿病大鼠有一定的改善作用,可为七箭方的优化提供实验依据。
OBJECTIVE: To observe the effect of different compatibility methods of traditional Chinese medicine on diabetic rats, and to explore the factors affecting the efficacy of Qijianfang. METHODS: Diabetic rat models were established by high-fat diet plus low-dose streptozotocin injection. Five different combinations of drugs were designed with a modified uniform design method. Different combination drugs were administered for 30 days to detect blood glucose and triglycerides. Cholesterol, serum creatinine, urinary creatinine, urinary albumin, creatinine clearance, and regression analysis of experimental results. Results: Blood glucose, triglyceride, cholesterol, and urinary albumin were significantly lower in experimental group 1-5 than in model group (P<0.05 or P<0.01); Creatinine clearance rate in experimental group 3-5 compared with model group The increase was statistically significant (P<0.05 or P<0.01). According to the regression model, the influence factors on different indicators were analyzed. The results showed that berberine had the most significant influence on blood glucose; the most important factors affecting triglyceride were the trichosanthin polysaccharides, berberine, and flavonoids; they had an important effect on cholesterol. The factors were Coptis berberine, Trichosanthus polynerum polysaccharide, Podophyllum flavone flavone. The major influencing factor for the creatinine clearance rate was notoginsenoside. The most important influencing factors for urinary microalbumin were Pleurotus flavone, Coptis berberine, Notoginsenosides, trichosanthin polysaccharides. Conclusion: The improved uniform design method is simple and feasible, it has a certain improvement effect on diabetic rats, and can provide experimental basis for the optimization of Qijianfang.
