目的在细胞实验中探讨靶向性人端粒酶逆转录酶(hTERT)启动子携带人类钠碘转运体(hNIS)基因能否提高胶质瘤的摄碘能力。方法检测在hTERT启动子引导下表达hNIS基因的腺病毒,使用该腺病毒转染U87和U251细胞系,然后使用γ记数仪检测转染后细胞的摄碘能力。结果实验组肿瘤细胞在转染Ad-hTERT-hNIS后,相对于对照组细胞摄碘率提高约30~40倍。结论实验证明hTERT启动子能引导胶质瘤细胞系表现出肿瘤特异性的碘摄取。
OBJECTIVE: To investigate whether the hTERT promoter hNIS gene can enhance the iodine uptake capacity of glioma cells in cell experiments. Methods The adenovirus expressing hNIS gene under the guidance of hTERT promoter was detected. U87 and U251 cell lines were transfected with the adenovirus. Then the radioactivity of the transfected cells was detected by gamma counter. Results Compared with the control group, the uptake of iodine in the experimental group of tumor cells increased about 30- to 40-fold after transfected with Ad-hTERT-hNIS. Conclusion Experiments show that hTERT promoter can guide glioma cell lines to show tumor-specific iodine uptake.
