目的 :比较黄芪和非创伤性肢体缺血两种预处理方法对大鼠离体心肌再灌注损伤的保护作用。方法 :采用雄性大鼠 ,在Langendorff装置上对离体心脏进行灌流。预灌 15min后 ,缺血 15min ,随后 15min再灌注。观察先用含黄芪注射液 ( 10mg/L)预灌在相应点分别测定冠脉流出液和心肌匀浆中SOD的活性和MDA含量 ,同时记录心肌细胞的单相动作电位 (MAP)和心肌收缩张力曲线。结果 :黄芪预处理和非创伤性肢体缺血预处理能显著降低再灌注心律失常发生率 ,显著降低心肌组织中MDA含量 ,提高SOD活性 ,稳定心肌细胞的膜电位和肌张力。结论 :黄芪预处理对大鼠离体心肌再灌注损伤有明显的保护作用 ,可能是通过增强心肌的抗氧化能力、稳定心肌的膜相结构等途径而实现的。
Objective : To compare the protective effects of two kinds of preconditioning methods of astragalus and non-traumatic limb ischemia on isolated myocardial reperfusion injury in rats. Methods : Male rats were used to perfuse isolated hearts on a Langendorff apparatus. 15 min after pre-irrigation, 15 min ischemia, followed by 15 min reperfusion. Observe the activity of SOD and MDA content in coronary effluent and myocardial homogenate respectively by pre-irrigation with astragalus injection (10mg/L) at the corresponding points. At the same time, record the single-phase action potential (MAP) and myocardial contraction of myocardial cells. Tension curve. Results: Pretreatment with astragalus and non-traumatic limb ischemic preconditioning can significantly reduce the incidence of reperfusion arrhythmia, significantly reduce the content of MDA in myocardial tissue, increase the activity of SOD, and stabilize the membrane potential and muscle tone of myocardial cells. Conclusion: Astragalus membranaceus preconditioning has significant protective effect on isolated myocardial reperfusion injury in rats, which may be achieved by enhancing myocardial antioxidation ability and stabilizing the membrane structure of myocardium.
