本实验取胎龄为23~24d的胎兔大脑皮层组织,分别以iloprost液及生理盐水为保存液,在4℃保存前后制成细胞悬液,不同浓度碘化丙啶染色后经流式细胞分析和台盼蓝排斥试验测定细胞活性。结果表明,iloprost液可以对兔胎脑皮质提供良好的保护作用,但随保存时间延长,细胞活性仍不可避免地下降,提示应进一步深入研究。最后,本研究还表明不同浓度碘化丙啶染色经流式细胞分析用于细胞活性测定较台盼蓝排斥试验更敏感、准确。
In this study fetus rabbit fetus from gestational age of 23 ~ 24d cortex tissue, respectively, iloprost solution and saline as preservation solution, before and after preservation at 4 ℃ made of cell suspension, propidium iodide different concentrations stained by flow cytometry Analysis and trypan blue exclusion assay determine cell viability. The results showed that iloprost solution can provide good protection to the fetal cerebral cortex, but with the prolongation of storage time, cell activity still inevitably declined, suggesting further study. Finally, the present study also showed that propidium iodide staining at different concentrations was more sensitive and accurate than trypan blue exclusion assay by flow cytometry for cell viability assays.
