背景:肺间质纤维化病理上可见以成纤维细胞(fibroblast,Fb)为主的大量间质细胞的增生及胶原分泌的增多,氟伐他汀对Fb等多种细胞具有增殖抑制作用。目的:观察氟伐他汀对体外培养的大鼠肺Fb增殖的抑制作用和对博来霉素诱发的大鼠肺纤维化及肺通气功能的影响。单位:解放军第四军医大学西京医院呼吸内科;解放军第四军医大学基础部病理学教研室;解放军第四军医大学西京医院骨科研究所。设计:随机对照实验研究。材料:本实验于200101/12在解放军第四军医大学西京医院呼吸内科实验室完成。实验选用健康雄性一级SD大鼠31只。干预:取1只大鼠,摘取其肺组织,体外培养大鼠肺Fb,加入氟伐他汀,MTT法观察对培养细胞生长曲线的影响及不同浓度(0,1×10-7,1×10-6,1×10-5,1×10-4,1×10-3,1×10-2mol/L,氟伐他汀0mol/L为空白对照组)的氟伐他汀对培养细胞的抑制作用;计数法观察氟伐他汀对大鼠肺Fb分裂指数的影响;羟脯氨酸比色法检测氟伐他汀对培养的大鼠肺Fb胶原分泌的影响。随机将另30只大鼠分为正常对照组、博来霉素组及氟伐他汀组。氟伐他汀组根据治疗开始时间为模型制备后第1,15天和给药剂量为20,10mg/kg(高、低剂量)分为4个治疗组:第1天高、低剂量组,第15天高、低剂量组。28d后处死动物,肺组织作HE、Masson和苦味酸天?

OBJECTIVE: To observe the inhibitory effect of fluvastatin on Fib proliferation in vitro and the effects of bleomycin-induced pulmonary fibrosis and pulmonary ventilation in rats.
