催产素(oxytocin,OXT)在动物分娩、维持哺乳方面具有重要作用,近年来发现OXT对能量代谢亦具有积极作用。尽管OXT在小鼠、大鼠、猕猴等动物脑部有广泛的分布表达,但OXT在绵羊下丘脑的表达部位和神经投射通路尚无报道,成为限制研究其生物学功能的瓶颈之一。本试验以绵羊下丘脑为主要研究材料,运用免疫荧光技术明确了OXT神经元在下丘脑的表达分布。结果显示,OXT神经元集中分布在绵羊下丘脑室旁核(paraventricular nucleus,PVN)区域,在视上核(supraoptic nucleus,SON)有零星表达,且均具有较为明晰神经元胞体及纤维结构,这种分布与小鼠的表达部位呈现高度一致。上述结果为进一步研究OXT在绵羊能量代谢方面发挥的生物学作用奠定了免疫学基础。
Oxytocin (OXT) plays an important role in animal delivery and breastfeeding. In recent years, OXT has also been shown to have a positive effect on energy metabolism. Although OXT is widely distributed in the brains of mice, rats and macaques, the expression of OXT in the hypothalamus of the sheep has not been reported yet, which has become one of the bottlenecks restricting the study of its biological functions. In this study, sheep hypothalamus as the main research material, the use of immunofluorescence technology to clear OXT neurons in the hypothalamus expression and distribution. The results showed that OXT neurons were concentrated in the paraventricular nucleus (PVN) region of sheep and sporadically expressed in the supraoptic nucleus (SON), all of which had distinct neuronal somatic cells and fibrous structures Species distribution and expression of the mouse site showed a high degree of consistency. These results laid the foundation of immunology for further study on the biological role of OXT in sheep energy metabolism.
