目的 寻求一种顺铂肾毒性的防护剂 ,给临床化疗提供手段和依据。方法 对C 57BL/ 6小鼠在给予顺铂( 1 4mg/kg)前后 2 4h分别给予醋酸锌 ( 2 0mg/kg) ,4d后通过电镜及光镜观察其肾脏的形态学改变 ,并测定血清尿素氮(BUN)及肌酐 (Cr) ,观察肾脏的功能改变。结果 C 57BL/ 6小鼠给顺铂前预先给锌可使顺铂对贤脏的损害明显减轻 ,锌对顺铂的抗癌效果无明显影响 (单用顺铂组抑瘤率 52 .91 % ,锌与顺铂合用组抑瘤率 50 .97% ,P >0 .0 5)。结论 锌可作为顺铂肾毒性的有效防护剂 ,并有希望用于临床。
Objective To seek a cisplatin nephrotoxic protective agent, to provide means and basis for clinical chemotherapy. Methods Zinc acetate (20 mg / kg) was given to C57BL / 6 mice 24 hours before and after cisplatin (14mg / kg) respectively. Morphological changes of kidney were observed by electron microscope and light microscope after 4 days. Serum Blood urea nitrogen (BUN) and creatinine (Cr) were observed in renal function changes. Results C 57BL / 6 mice given cisplatin pre-given zinc can significantly reduce the harm of cisplatin on the dirty, zinc on the anticancer effect of cisplatin had no significant effect (single cisplatin group tumor inhibition rate of 52.91% , Zinc and cisplatin combination group inhibition rate of 50.97%, P> 0.05). Conclusion Zinc can be used as an effective protective agent of cisplatin nephrotoxicity, and hopefully for clinical use.
