目的研究阿魏胶囊对载脂蛋白A4(Apo-A4)基因转录水平的影响。方法将大鼠随机分为正常组、模型组和阿魏胶囊预防中、高剂量组及阿魏胶囊治疗中、高剂量组,共6个组。除正常组仅用普通饲料喂养外,其余各组每天灌以高脂乳剂2mL。其中预防2组造模当天灌服阿魏胶囊药物混悬剂4周;治疗2组连续造模4周后,待造模成功,再灌服阿魏胶囊药物混悬剂4周。各组取血液后采用逆转录-聚合酶链反应(RT-PCR)对Apo-A4进行检测,观察其表达水平。结果RT-PCR凝胶成像系统结果分析表明,大鼠Apo-A4基因的转录水平正常组与模型组相比,差异有统计学意义(P<0.05);模型组与预防中、高剂量组及治疗中、高剂量组相比差异有统计学意义(P<0.01)。治疗中剂量组和预防中剂量组的转录水平都略高于治疗高剂量组和预防高剂量组。结论阿魏胶囊明显地提高或稳定Apo-A4基因的转录水平。
Aim To study the effect of awe capsule on the transcription of apolipoprotein A4 (Apo-A4) gene. Methods The rats were randomly divided into six groups: normal group, model group, and Ewe capsule for the prevention of middle and high dose group and Ewei capsule for medium and high dose group. In addition to the normal group fed only ordinary feed, the remaining groups were infused with high-fat emulsion 2mL daily. The prevention of two groups of modeling the day of avalais capsule drug suspension for 4 weeks; treatment of two groups after 4 consecutive weeks of modeling, to be successful modeling, and then remitted the capsule Wei Wei capsule suspension for 4 weeks. Apo-A4 was detected by reverse transcription-polymerase chain reaction (RT-PCR) after the blood was taken from each group, and the expression level was observed. Results The results of RT-PCR gel imaging system analysis showed that there was a significant difference in the transcription level of Apo-A4 gene between normal group and model group (P <0.05) There was significant difference between the middle and high dose treatment groups (P <0.01). The transcript levels in the middle-dose and middle-dose groups were slightly higher than those in the high-dose and high-dose groups. Conclusion Ferulic capsules significantly increase or stabilize the transcription level of Apo-A4 gene.
